Research reference
Tirzepatide (development code LY3298176) is a fatty-acid-modified peptide with dual GIP and GLP-1 receptor agonist activity, built on a GIP peptide backbone conjugated to a C20 fatty acid moiety. Binding affinity and cAMP-accumulation potency have been characterized in HEK 293 cells transfected with the human GIP and GLP-1 receptors. Signaling studies describe greater receptor engagement at GIP than GLP-1, and bias at the GLP-1 receptor favoring cAMP generation over β-arrestin recruitment, with weaker receptor internalization than native GLP-1.
