Tesamorelin is a synthetic 44-amino-acid analogue of growth hormone-releasing hormone, structurally trans-3-hexenoyl-GHRH(1–44)NH₂. The N-terminal hexenoyl modification protects the molecule from dipeptidyl peptidase-4 cleavage, conferring greater stability than native GHRH, whose plasma half-life is measured in minutes. It acts as a selective agonist at the GHRH receptor on anterior pituitary somatotrophs, and published work reports augmented basal and pulsatile GH secretion with increased IGF-I. Research use only.
